Two things to try:
First, try running software-center from a terminal window and see if there
are any useful error messages.
If that doesn't help, try reinstalling software-center:
sudo apt-get purge software-center
sudo apt-get install xubuntu-desktop
You can also try using synaptic as an alternative to software-center.
