Do you have windows ?
Have you installed Ubuntu as dual boot or through wubi ?

That would indicate that the indicator / icon is working.

try using alsamixer

-$ alsamixer

see what volume your mic has been set too, by default it would be at
zero, change it higher.

Regarding skype
using the preferences / options menu to change you settings and see
what works by making a test call.
