Work out why first. There is already a distro to handle just about every
special purpose niche and a glut of general purpose distros:

You will want to be familiar with filesystem layout, shell scripting,
compiling, security, a packaging system and lots more. For a general
purpose/desktop distro it would be best to start with something
established such as Ubuntu, Debian, Fedora etc.

Some of the so-called distros are just one of the major distros with a
different menu and desktop theme and not much else.

If you want to build a system to understand how the bits go together
something like http://www.linuxfromscratch.org/ might be the way to go.

There are also distros to build distros
http://www.rocklinux.org/wiki/Main_Page

Ubuntu can be so polished it might be hard to see what is going on
behind the scenes. Perhaps Gentoo or Slackware might be worth a look.

There are some useful small special purpose distros such as puppylinux
that might be worth trying. They have less complexity.

Another suggestion is instead of going alone why not learn from the best
and help out an established distro such as Ubuntu?
