resolv.conf will be overwritten the next time that you renew your DHCP
lease unless you exclude acquiring DNS servers from your DHCP lease
request.

Check the DNS settings that the DHCP server is leasing out since
adding DNS servers manually solves your problem.

I assume that Skype uses IP addresses and therefore does not use DNS.

(If you want to manage your NIC with NM but want to set up your NIC in
/e/n/i, change the "ifupdown" section of
/etc/NetworkManager/nm-system-settings.conf to "managed=true".)
