I've heard good things about maildir, but I use mbox myself.
I've got a decently powerful laptop, and some of my
mailboxes contain tens of thousands of messages; the
ubu-users mailbox, for instance, contains just under 16,000
messages. It takes quite a while to load in mutt -- maybe 15
seconds or so (though I've not timed it). Subsequent loads
don't take long. Saving a message to ubu-users takes no
time; I gather that appending a message to an mbox file is
a constant-time operation.
