It's up to you. I found GUI programming very educational when I first learned
it (that was back in Amiga days though, and the Amiga GUI was quite well
thought-out for the time).
In this case, I would go with glade, though. RAD tools are very useful for
letting you just get stuff done, and when you're learning, you don't want
boring details distracting you from what you're trying to understand.
Concentrate on coding your own stuff well, learn what you need to from gui
programming as you go, and avoid the internals of pygtk as much as you can;
it doesn't seem that well-designed anyway, so it's probably not something you
want to learn techniques from.
