Which one?  The procedures are completely different.

You won't want to change anything but /casper/filesystem.cloop.  You are
free to replace that file with any cloop-compressed ext2 filesystem image
you like, created by any means which is convenient for you.  You can use
debootstrap to create a starting point, and chroot into it to make further
changes.
