No, it won't, except the file system on your portable HDD is damaged. You should edit the grub menu with your correct "boot" device (portable HDD), if you are not sure, you can "c" or "ctrl-c" into the grub shell, try the command of "ls", you will get all of the device number. try to identify your real "boot" device.
