no, as all gconf settings it takes effect immediately.

the only tooltips left in lucid are actually the ones on the panel items
(if you add apps or applets to the panel) which this setting is supposed
to switch off. 

the rest of the (default) system should actually be tooltip clean beyond
some desired places like the weather icon and network manager.

if there are any tooltips seen beyond that you might have found a
bug ... (the desktop experience team did put some effort into tooltip
handling during lucid afaik)

btw, i just checked, at least for me it switches on/off tooltips on the
panel as desired
