Unfortunately I lost my Debian testing by an accident, but I can't remember such a time-lag. Anyway, I'm using Arch Linux with Xfce and Thunar now. If I click the icon to open it, it's directly opened with the click. No delay here.
$ uname -r
3.1.5-1-ARCH
$ thunar --version
Thunar 1.2.3 (Xfce 4.8)
64 bit AMD Athlon dual core BE-2350 2.1 GHz 4 GB RAM

PS: FILE SYSTEM EXT3 INSTEAD OF EXT4 ;), but I don't think this is the cause that it opens like a bat out of hell here.
PPS: I didn't open it for the first time, dunno how slow or fast this would be, the drive is an averaged SATA with an averaged cach. Currently I'm not running much apps, perhaps Thunar is stored in the main memory.
