You might try the following as a work-around:

Open your dvd directory (created by dvdbackup) with pgcedit (a Windows program)
Go to the Title menu, and select 'rebuild all time maps of dvd'
Save
Move the PgcEdit_backup directory in VIDEO_TS to someplace else
Re-try your previously failed mkisofs

Bug description:
  Making a DVD Video ISO from the Porcupine Tree "Anesthetize" DVD
  Audio/Video the same way as many other successful ISOs from either
  files created by "dvdbackup --mirror" or automounted consistently
  fails with "Implementation botch. Video pad for file VIDEO_TS.BUP is
  -1". The DVD plays fine in any player I've tried.

  Using "-no-pad" makes no difference.
