Does it work if you use the full path for the file instead of prefixing the filename with "~"?
I was able to use the -i option "-i /var/downloads/s-u/ubuntu.com/ubuntu-12.04-desktop-amd64.iso" with version 0.2.23.

Bug description:
  Binary package hint: usb-creator

  On Karmic.

  I can't add any iso file to the menu nor dragging it with the mouse nor choosing it through the menu.
  If run from the command-line:

  
  streinsacu at roncalle:~$ usb-creator-gtk --ISO=~/Downloads/ubuntu-9.10-alternate-i386.iso 
  Usage: usb-creator-gtk [options]

  usb-creator-gtk: error: no such option: --ISO
  streinsacu at roncalle:~$ usb-creator-gtk --iso=~/Downloads/ubuntu-9.10-alternate-i386.iso 
  isoinfo: No such file or directory. Cannot open '~/Downloads/ubuntu-9.10-alternate-i386.iso'. Cannot open SCSI driver.
